Is it ok if I install guides sitting on the front face frame and screwed into the side screw holes all the way back, but not sitting on anything in the rear or a rear support of any kind?

I have done that more times than not. When I build face frame cabinets, the sides of the cabinets where drawers will be, are flush with the inside of the face frame, just for that reason.
If the cabinet sides are plywood, yes. If the cabinet sides are solid lumber with the grain running up-down, the rear screws should really be able to slide.
I would at least apply a glue block at the back.
It should have a shoulder of some sort.
