I am making a 6" x 24" sign from scrap Boral. Do you think I could insert plastic wall anchors along the edge and hang the sign similar to the one in the photograph?

One other option is to place a threaded rod all the way through it and use nuts on the ends and then hang it that way. What are your thoughts?

I'd probably opt to epoxy a threaded insert and then a threaded eye bolt. Dont know that Id trust plastic anchors over the long haul.

Lag bolt eye or hook.

I would use RedHeads (threaded wedge anchor). If you have not used them before, just remember they tighten up when you PULL them, not when you pound them in.
EDIT - - I made a recommendation for the brick. If going into wood, then Lag or E-Z LOK flanged insert and eye bolt.

Boral is a great product for exterior use.
Totally impervious to weather.
Use same screw pilot hole chart for hardwood.
Round eye hook at least 1" long will suffice.
